Kawasaki Frontale Youth Academy (2010–2014)
Itakura developed his defensive fundamentals within the highly respected youth academy of Kawasaki Frontale. He consistently captained his age-group teams, showcasing early signs of the leadership qualities that would define his professional career.

Ko Itakura commenced his professional journey with Kawasaki Frontale, making his senior debut in 2015. After a successful loan spell at Vegalta Sendai in 2018, where he gained valuable first-team minutes, he attracted the attention of Premier League heavyweights Manchester City. He signed for City in January 2019, embarking on his highly anticipated European adventure.

Due to work permit regulations, Itakura was immediately loaned to FC Groningen in the Dutch Eredivisie. Over two and a half seasons, he adapted perfectly to European football, becoming a defensive stalwart and a fan favorite. In 2021, he joined German side Schalke 04 on loan, where his exceptional performances were instrumental in helping the historic club secure promotion back to the Bundesliga.

Seeking a permanent home, Itakura signed with Borussia Mönchengladbach in the summer of 2022. During his three-year tenure with the Foals, he proved himself against some of the world's best attackers, showcasing excellent reading of the game and strength in ground duels. His consistent form made him one of the standout center-backs in the German top flight.

In August 2025, Itakura completed a major transfer to Ajax Amsterdam, signing a contract until 2029. Becoming the first Japanese player in the club's illustrious history, he quickly assumed a leadership role in their young defensive line. Throughout the 2025-2026 season, he has been an integral part of Ajax's squad in both domestic and European competitions, offering vital stability and experience.

Internationally, Itakura has been a stalwart for Japan. He represented his country at the 2020 Tokyo Olympics and the 2022 FIFA World Cup, where he famously provided a crucial assist against Germany. Heading into the 2026 World Cup, he is considered one of the indispensable pillars of the Japanese defense.
